Protein synthesizing activity and membrane transport were examined in fresh and cryopreserved isolated rat liver mitochondria. In the presence of 0.6, 1.2, and 1.8 M final concentrations of dimethyl sulfoxide (Me2SO), both metabolic parameters were considerably inhibited in the fresh samples and even more inhibited in the cryopreserved specimens. However, simple exposure to this penetrating cryoprotectant, followed by its subsequent removal by washing, did not seem to affect significantly the examined functions. When different freeze-thaw regimes were investigated, it was observed that optimal recovery of protein synthesis and membrane transport functions were obtained when fast freezing took place in the absence of Me2SO.  相似文献

We have isolated a potent proteinaceous chemoattractant from aqueous washes of earthworm (Lumbricus terrestris) for garter snakes (Thamnophis sirtalis) by means of a covalent chromatography. It contained free sulfhydryl groups and showed an apparent mass of 20 kDa. The chemoattractive activity of this protein could be destroyed by heating as well as by proteolysis. Its activity could also be reversibly blocked by mixed disulfide formation with dithiodipyridine, suggesting that the free sulfhydryl(s) was essential for its function as a chemoattractant. This bioactive material had a tendency to form intermolecular crosslinked aggregates during isolation, if reducing agents were not included. Some of the high-molecular-weight aggregates cochromatographed with earthworm cuticle collagen on Ultragel AcA 34 or 44 columns. In contrast to an earlier report by D. M. Kirschenbaum, N. Schulman, and M. Halpern [1986) Proc. Natl. Acad. Sci. USA 83, 1213-1216) the purified earthworm collagen showed no chemoattractive activity to garter snakes.  相似文献

The biological and immunological activities of Staphylococcal enterotoxin B are stable in pH 2.0 approximately 11.0, and also resistant to proteolytic enzyme (trypsin, pepsin) digestion for 3 approximate 4 hours. Therefore, the toxin could be purified with trypsin digest and then absorbed on kaolin and Kieselgel, followed by eluting the different pH buffer solutions. Ultravoilet photoelectron spectroscopy has been employed to exmamin the valence electronic structure of 5-fluorouracil, 5-chlorouracil, 5-bromouracil, and 5-iodouracil. Photoelectron bands associted with the three highest π orbitals and the two oxygen atom lone-pair orbitals were assigned by a comparison to similar bands observed in the photoelectron spectrum of uraciul. Bands arising from the halogen atom lone-pair orbitals were assigned by comparing the present results with photoelectron spectra measured for halobenzenes, and by considering the linear dependence of halogen atom lone-pair ionization potentials upon halogen atom electronegativities. The present spectroscopic results have been compared with results from studies of association constants of 5-halouracil–adenine complexes. This examination in dicates that the complex association constants incresase as the ionization potentials of the highest occupied π orbital and the halogen atom lone-pair orbitals of th halouracils decrease.  相似文献

Horses suffer from a respiratory condition, similar to human allergic asthma, that is characterized by severe dyspnea, wheezing, coughing, and mucus production. Mediator substances released during the allergic reaction may contract airways and pulmonary vasculature. Nothing is known of the effects of autacoids and other vasoactive substances on equine pulmonary vessels. Therefore, spiral strips of equine pulmonary vein were prepared in vitro and the effects of histamine (H), 5-hydroxytryptamine (5HT), bradykinin (BK), carbachol (Carb), and phenylephrine (phen) were studied. The order of contractile effectiveness for the agonists on the vein was found to be 5HT greater than H greater than Bk greater than Phen greater than Carb, although H consistently produced the greatest maximal effects. H1-receptors appeared to mediate H contractions while H2-receptors had no measurable effect. 5HT responses were mediated directly by 'D-type' smooth muscle receptors. Bk produced contractions but of a lesser magnitude than either H or 5HT. Varying degrees of tachyphylaxis were observed for each agent. alpha-Adrenergic receptor stimulation by Phen initiated low-magnitude contractions whereas Carb exhibited virtually no activity on the pulmonary vein. Contractile responses of pulmonary veins to various spasmogens may contribute to the equine asthmatic response by raising vascular hydrostatic pressure, thereby enhancing edema formation.  相似文献

Following intravenous administration of the cholinesterase reactivator HS-6 (30 mg/kg), blood pressure fell (up to 50 mmHg) and maximal blood levels of HS-6 reached 242 microgram/ml. HS-6 attenuated the pressor response resulting from carotid occlusion and the depressor effect of vagal stimulation. Doses of HS-6 below those used to protect against soman in different animal species (10--30 mumol/kg) progressively blocked the ganglion-stimulating effects of nicotine and dimethylphenylpiperazinium but not the pressor effect following adrenaline, a pattern similar to that produced by hexamethonium but only 1/84 as potent. HS-6, like hexamethonium and mecamylamine, progressively blocked the contraction of the nictitating membrane of the cat resulting from preganglionic stimulation. The results indicate that HS-6 possesses ganglion-blocking properties at doses likely to be used in the protection against soman poisoning. The ganglion-blocking properties of the drug may be a factor in the beneficial effects of HS-6.  相似文献

Since ovarian follicles appear to be randomly oriented with respect to the plane of the section, the method of sectioning and examining follicles at their maximum diameter described here allows direct comparison between oocyte populations of women and small differences can be detected. Ribonuclease (EC 2.7.7.17) activity in the obligate halophyte Suaeda maritima (L.) Dum. var. macrocarpa Moq. was studied in relation to salinity (increasing concentrations of NaCl) of incubation and growth media. In vitro, the addition of 50 to 400 m M NaCl did not affect ribonuclease activity. This result, which was also found for Phaseolus vulgaris , indicates that the hydrolase is insensitive to high saline concentrations. The subcellular distribution of RNase activity did not change significantly with the salinity of the medium or with the age of the plant. The microsomal ribonuclease activity expressed on a fresh weight basis represented in every case less than 6% of the total activity. After 23 days of culture, the absence of salt stimulated the activity of soluble ribonuclease in aerial parts of Suaeda ; inversely, the capacity of the enzyme was lower under optimal saline conditions (130 m M NaCl). This was also evidenced by transfer of whole plants from a non-saline to a saline medium. Such a saline shock caused a decrease followed by a stabilization of the capacity of ribonuclease from Suaeda . The influx of NaCl in the tissues lowered the activity of the hydrolase.  相似文献

A study was made of the chronic effect of pesticides, within a wide range of doses, combined with whole-body gamma-irradiation, on the content of carbohydrate-containing proteins of rat liver. The extent to which disturbances in the carbohydrate-containing protein metabolism were manifest was shown to depend upon dose and time of action of the damaging factors. As determined by certain parameters, the combined effect of pesticides and gamma-radiation at the biochemical level was additive, and it was more than additive as estimated by a number of other tests.  相似文献
